High throughput multiparameter analysis of single mitochondria
Mitochondria are the energy factory of cells and play an important role in the evolution of eukaryotic cells. In addition, mitochondria are also closely related to intracellular signal transduction, such as autophagy, cell death, natural immunity and so on. A variety of complex human diseases are caused by mitochondrial dysfunction, including neurodegenerative diseases and metabolic disorders. Therefore, mitochondrial research is of great significance for basic life science research. The structure of mitochondria is small, and there is great heterogeneity (heterogeneity) between different mitochondria. Its diameter is generally 0.5-1.0 μ M length is 1.5-3.0 μ M scattering light intensity is equivalent to that of 100-200 nano polystyrene microspheres, and the content of biochemical substances is low. The detection of single mitochondria has always been a challenging topic. So far, although flow cytometry plays a leading role in the analysis of single mitochondria, such as the purity of mitochondria, membrane potential and morphological changes induced by calcium ions by flow cytometry, Reactive oxygen species (ROS) were detected. However, mitochondrial analysis is only limited to biochemical parameters that can be stained by high brightness fluorescence. The significant improvement of the sensitivity of nano flow detector will provide advanced characterization technology for single mitochondrial analysis.
1. Rapid characterization of mitochondrial purity and structural integrity
On the nano flow detection device, The brain and SYTO 62 were used to specifically label the cardiolipin on the mitochondrial inner membrane and the mitochondrial nucleic acid (mitochondrial DNA) in the matrix. The purity of mitochondria and the integrity of inner membrane structure could be investigated by scattering of single mitochondria and simultaneous detection of endocardial cardiolipin and mitochondrial DNA Simultaneous detection of (porin) protein and CYTS protein in membrane gap can quickly characterize the integrity of outer membrane and function of mitochondria.
