血管内皮生长因子(VEGF)最初被称为血管通透性因子(VPF)，是一个刺激血管发生和血管新生的蛋白质家族。VEGF家族包括五个成员：VEGF-A、VEGF-B、VEGF-C、VEGF-D和PGF（胎盘生长因子）。作为第一个被发现的VEGF家族成员，VEGF-A常被简称为VEGF。VEGF-A基因由8个外显子组成，选择性剪接产生10种以上亚型，包括VEGF 121、VEGF 145、VEGF 165、VEGF 183、VEGF 189和VEGF 206等。VEGF165和VEGF121是在大多数细胞和组织中表达的主要亚型。VEGF165是一种45KDa同源二聚体糖蛋白，对肝素具有中等亲和力。相反，VEGF121不与肝素结合。VEGF特异性结合酪氨酸激酶受体VEGFR-1(Flt-1)和VEGFR-2(Flk-1)，激活下游信号通路，导致内皮细胞存活、增殖、迁移和血管通透性。VEGF通过增强血管通透性或招募内皮祖细胞到新生血管形成部位远处位点，在病理性血管生成中起着核心作用。随着对VEGF促进病理性血管生成和肿瘤转移的深入了解，在多种临床模型中，阻断VEGF/VEGFR轴抑制血管生成和肿瘤生长已成为合理的研究方向。
