I now have some further information to share with you in regards to the anti-Calpain 1 antibody - Domain IV (ab39170).
The immunogen used to raise this antibody is taken from thecarboxyterminal area of Calpain-1. As proteolytic processing of Calpain-1 occurs at the amino end, the antibody is likely to be able to detect both thezymogen and cleaved enzyme. It is therefore possible that you have been picking up both forms in your Western blot.
