In the world of fluorescent dyes, ROX is often used to refer to ROX derivatives with a 5/6 carboxylic acid structure. This carboxylic acid structure serves as a flexible bridge connecting fluorescent dyes to target biomolecules, enabling the activation and labeling of fluorescent dyes. NHS activated ester, an activating group in the biomarker reaction, is chemically named N-hydroxysuccinimide ester (SE). It activates the carboxyl group in the dye molecule, enabling the dye to react accurately with the amino groups (mainly primary amines) on the target biomolecule, generating stable amide bonds, thereby firmly labeling the dye molecule on the biomolecule. This reaction ensures both the accuracy of labeling and the stable connection between dye molecules and biomolecules.
